Notre équipe travaille sur l'impact des récepteurs nucléaires et de leurs cofacteurs sur le vieillissement métabolique qui mène à l'obésité, au diabète de type 2 et à l'athérosclérose. Par des techniques de biologie moléculaire et cellulaire de fine pointe sur les liaisons spatio-temporelles des facteurs transcriptionnels et de leurs cofacteurs nucléaires, notre focus porte sur la découverte de nouveaux gènes liés au métabolisme dans des tissus cibles (adipeux, foie, pancréas) et au contrôle de leur expression. Notre objectif principal est la découverte et le développement de nouvelles cibles thérapeutiques contre les maladies métaboliques et le vieillissement prématuré.
Description des travaux
Une hypothèse centrale au coeur des recherches de notre laboratoire est que le vieillissement métabolique est causé par un débalancement des cofacteurs nucléaires, ce qui mènerait à une activité transcriptionnelle anormale et le développement de maladies comme l'obésité et le diabète de type 2. Nous utilisons plusieurs modèles de vieillissement (nématodes, souris, rats, humains) et tentons de les mimer en culture cellulaire. Dans chacun des cas, nous voulons établir l'activité et les niveaux de chacun des cofacteurs dans plusieurs tissus métaboliques (e.g. adipeux, foie, pancréas, muscle, coeur). Cette approche matricielle unique apporte une puissance expérimentale incomparable et est complétée par notre nouvelle plateforme génomique de « differential display » qui permet la découverte de nouveaux gènes d'importance. Par la suite, nous validons ces concepts moléculaires chez des souris transgéniques et par des approches nutritionnelles et/ou pharmacologiques. Ces recherches visent la découverte et le développement de nouvelles cibles thérapeutiques contre les maladies métaboliques et le vieillissement prématuré.
